9、Programmed cell death drug therapy research
This major aims at the development of programmed cell death drugs. The autophagy and programmed cell death in human cancer cells are mainly discussed in terms of morphological characteristics, key molecular processing mechanisms, signal transduction, apoptosis and necrosis, their role in pathophysiology and their research methods. Programmed cell death (PCD) serves fundamental functions during bothplantandanimaltissue development.Apoptosisandautophagyare both forms of programmed cell death, butnecrosiswas long seen as a non-physiological process that occurs as a result of infection or injury.Necrosisis the death of a cell caused by external factors such as trauma or infection and occurs in several different forms. Recently a form of programmed necrosis, callednecroptosis, has been recognized as an alternate form of programmed cell death. It is hypothesized that necroptosis can serve as a cell-death backup to apoptosis when the apoptosis signaling is blocked by endogenous or exogenous factors such as viruses or mutations. Most recently, other types of regulated necrosis have been discovered as well, which share several signaling events with necroptosis and apoptosis.
Supervisor:Prof. Cheng-yun Jin
10、Anti-viral medicine research
Anti-viral medicine research group is now working on anti respiratory medicines, including anti-influenza drugs and anti-respiratory syncytial virus drugs. We have closely integrated with the Department of medicinal chemistry and natural medicine of the school of pharmacy, Zhengzhou University. They provide compounds, and we are primarily responsible for the evaluation and mechanism of antiviral medicines. Our research group found that lactone compounds have anti-viral respiratory function, we analyze the mechanism of antiviral medicines of lactone compounds has two parts: first one is the direct effect on the virus surface protein, second one is enhance antiviral immunity in virus binding with the receptor, such as stimulating secretory expression of surface receptor RIG-I and TLR. We welcome you to join our group in the study of antiviral medicines.
Supervisor:Dr. Zhenya Wang
11、Antituberculosis drug research
Tuberculosis is a chronic contagious disease caused by a single agent,Mycobacterium Tuberculosis(Mtb), which has been a big threat to public heath worldwide. The only available vaccine for Mtb is BCG, whose efficacy is very limited. Besides that, more and more patients are infected by those stains with drug resistance to the current available drugs. Our research has been investing the membrane-lysis activity of ESAT6,a secreted toxin by Mtb. We found the mutation of some amino acid, which is critical to the toxicity of Mtb and membrane-lysis activity of ESAT6. We will continue to work on the possibility of ESAT6 as a vaccine candidate and potential new drug target for anti-Mtb therapy.
Supervisor:Prof. Qi Zhang
12、Anti-viral phosphonated nucleosides research
Substitution of nucleoside monophosphate with a bioisostere phosphonate nucleoside, can eliminate the rate-limiting monophosphorylation step in cell and increase hydrolysis stability of substrate as well, to greatly improve the bioactivity of substrate. Therefore development of novel phosphonated nucleosides is an effective efficient method to get potent antiviral drug candidate. In this project, based on the L-threosyl nucleoside scaffold, a series of novel phosphonated nucleosides will be synthesized by modification of the base and sugar moiety. Their biological activity will be systematically evaluated at cell assays, and the relationship of structure and activity will be studied as well. We would gain some insights into the mechanism of action of these compounds, and understand their intracellular activation and metabolism. Some prodrugs will also be synthesized with an aim to improving the efficiency of cell delivery and better understanding the bioavailability and pharmacokinetics. We will finally develop some novel phosphonated nucleosides as potential antiviral or antitumor agent candidates.
